To work in Australia, you will need authorisation (a valid visa) from the Department of Home Affairs, however you need a valid TFN so that you are not taxed at the highest marginal tax rates.

This means if you hold a valid work visa you can start work at anytime you want but without a valid TFN your employer must withhold tax from any payments that they make to you at the hightest rate of tax and they have no choice in the matter.
