As Title, if I’m using my wife’s car as uber eats delivery, do I need to swap the rego under my name in order to claim business deduction? Like insurance, rego fee, fuel, etc. which I paid for it. Just not under my name.

Yes - you can claim expenses.
When the car is someone else's vehicle, you can claim these also. You'll claim it as travel expenses rather than car expenses.
